<title>Hygiene Practice, Microbial Diversity and Detection of Pathogenic Bacteria in some Ready-To-Eat Foods of Rajshahi Metropolis</title>

RTE food has become an important public health issue and a great concern to everybody. This is due to widespread food borne diseases, due to the mushrooming of wayside food vendors who lack an adequate understanding of the basic food safety issues. In the present study different restaurants and street food vans of six hot spots of Rajshahi Metropolis were surveyed during March, 2015 to February, 2016. The survey was mainly conducted to ascertain the socioeconomic condition, hygiene and sanitation practice of food seller to reduce the contamination rate and &#13;
to p1&#1048606;otect public health. From the survey result it was observed that all RTE food vendors of Rajshahi Metropolis were male with the age range as below 25-50 years, illiterate to under H.S.C pass and have 5-20 years of experience in this profession. The majority of restaurants vendors earned 26000 to 30000 taka monthly. The most of the restaurant vendors had no knowledge about food hygiene and very much irregular for cleaning of sale surfaces (23.07%). Hand washing before and after serving was practiced by 23.07% vendors, 30.77 % used aprons and none of vendors covered hair with caps or used gloves while serving food. They used detergent for cleaning of utensils. None of the vendors have any training about food safety and not aware of food safety law. On the other hand, in case of street food vendors, it was observed that majority (100%) of street vendors were male. Most of the vendors (60%) were 36-45 years old and 64% were passed JSC and only 8% street vendors were illiterate. The majority (52%) of street vendors earned 11000 to 15000 taka monthly and majority (80%) had no knowledge about food hygiene. None of the street vendors used apron, gloves while selling food and did not covered food with polythene. ---

<title>Improvement of Potato (Solanum Tuberosum L.) Through in Vitro Culture</title>

<summary type="text">Improvement of Potato (Solanum Tuberosum L.) Through in Vitro Culture

In order to develop disease free potato seeds in vitro culture was established using meristems of potato cultivar, Diamant. Meristems were isolated from 25-30 days old field grown plants and cultured onto filter paper bridge in liquid MS medium. Among 29 media formulations medium with MS+0.1 mg/I KIN + 0.5 mg/I GA3 was found to be the best media formulation for primary establishment of meristem culture. Estabilished meristems were subcultured onto agar solidified medium. Among 22 medium formulations media with 0.5 !71911 BAP + 1.0 mg/I IBA was found to be the best for shoot proliferation. For all four potato cultivars viz. Diamant, Cardinal, Multa and Lalpakri sucrose was found to be the best carbon source and 3% sucrose was better than 6% sucrose for shoot proliferation. For adventitious root induction medium with MS+0.05 mg/1 IBA+ 0.05 mg/I GA3 was appropriate media composition among 16 media formulation and 3% sucrose was best among seven concentrations (10-70 g/1) of sucrose for root induction for all four cultivars of potato. Maximum in vitro tuberization was obtained in media supplemented with 8 mg/I KIN among six concentrations (2-12 mg/I) of BA and KIN. Medium contained 60 g/1 sucrose was found to. be the best for microtuber induction for all four cultivars among various concentration (10-70 g/1) of sucrose. Continuous photoperiods (24 hour) produced more microtubers than 0- and 8-hour photoperiods. &#13;
Comparative field evaluation of potato plants obtained from three propagule sources viz. tuber, micro tuber, and micro shoot indicate that tuber yield of tuber produced plants was higher than microculture (micro shoot, micro tuber) produced plants. Between microshoot and microtuber produced plants tuber yield of microshoot produced plants was higher than microtuber produced plants. But the tuber number of microculture produced plants was greater than the plants obtained from tuber. Disease reaction indicates that microculrture produced plants showed few contaminations by virus, bacteria and fungus than the tuber produced plants. &#13;
The results of the present study showed that disease free potato seeds could be developed through meristem culture which may help to increase potato production in Bangladesh

Different aspects of micropropagation through meristem culture for the production of virus free plants, callus induction, somatic embryogenesis followed by subsequent plant regeneration, in vitro bulblet formation and field evaluation of the in vitro regenerated plants were studied on five commercial cultivars of garlic (Allium sativum L.) viz., GC002, GC008, GC009, GC0025 and Indian variety. For micropropagation, shoot tip of field grown mature garlic bulbs of five cultivars were used for meristem isolation. Meristems were cultured on to filter paper bridge in liquid MS medium. Among the different media formulations MS + 1.0 mg/1 2ip + 0.5 mg/1 NAA was found to be the best for primary establishment of meristems and for maximum shoot proliferation. Protocol was also developed to induce in vitro bulblets from meristem derived shoot clumps and among various concentrations of sucrose, 12% sucrose was found to be the best for in vitro bulblet formation for all the four cultivars. Same result was also found in callus culture. Rooted plantlets with bulblets of four garlic cultivars were gradually acclimatized and successfully established in the field. Visual evaluation of the morphological traits of the meristem derived plants showed that all plants were normal and free from virus diseases. Substantial yield increase was observed from meristem derived plants over their source plants. Among the four cultivars cv. GC0025 was found to be highly responsive for in vitro shoot proliferation and bulblet formation.
